If the whole image has low signal, the problem is mostly in ionization efficiency, transmission or instrument state; if only certain regions are weak, it is mostly the sample itself (such as low molecular content, salt crystallization or morphological variation in that region) or scan-parameter drift with position.
Using standards or known high-signal regions as controls can quickly determine whether the problem is the source or the sample/region. This step avoids unnecessarily disassembling the ion source.
Check whether the sample is dehydrated, contaminated, has salt residues or is improperly stored; MALDI requires confirming whether matrix spraying is uniform and crystallization is good; DESI/LDPI/DPI require confirming surface cleanliness and spray/laser alignment.
