1. Device principle

AP-SMALDI enters the laser from the back of the sample, reducing surface charge accumulation and improving ion yield, and operates under ambient pressure (AP) to simplify sample transfer. It retains MALDI's high sensitivity to peptides, proteins and lipids, but sample preparation still requires matrix spraying and co-crystallization.

Ambient operation is its main convenience over vacuum MALDI: samples do not need to enter the vacuum chamber, making preparation and switching faster. But this path does not eliminate the matrix itself, and matrix peak interference in the low-mass region remains, which is the essential difference from matrix-free sources.

2. Differences from Neo-Source LDPI

Neo-Source LDPI also works under ambient pressure, but uses laser desorption plus photoionization, needs no organic matrix, has a low background above m/z≥70, and better suits small-molecule drug and metabolite imaging; its 2–3 μm resolution (among the best in the industry) reaches the single-cell/subcellular scale.

In short: AP-SMALDI 5AF is an ambient version of MALDI (still needs matrix), while LDPI is an ambient matrix-free source (no matrix background). If the focus is on small-molecule imaging with low background, LDPI fits better; if you prefer MALDI-class soft ionization and can accept a matrix, AP-SMALDI is an option.

3. Compatibility and selection

AP-SMALDI class sources require confirmation of specific instrument and analyzer support; Neo-Source LDPI adapts to Agilent, SCIEX and Thermo via external modular form, reusing the existing instrument.
